Optimization of Roche Liberase TM Research Grade in the Enzymatic Digestion of Human Adipose Tissue for the Isolation of Stem and Regenerative Cells

Adipose-derived stem cells are increasingly targeted as an autologous source of multipotent cells in regenerative medicine. Specifically, reconstructive lipotransfer procedures combine freshly harvested adipose tissue with ADSCs to improve vascularization and tissue grafting [1]. Furthermore, ADSCs offer a wide array of potential uses in tissue reengineering and cell therapies including wound healing, calvarial bone defect therapy, graft-versus-host disease treatment, and repair for ischemic damage [2]. Harvesting ADSCs requires the enzymatic digestion of adipose tissue to separate cellular components from adipose tissue. Methods for this procedure conventionally utilize Type I Worthington Collagenase which limits the therapeutic potential of ADSCs. ADSCs are harvested during liposuction and are also known as stromal vascular fraction cells. They have a heterogeneous population that includes adult cells, endothelial progenitor cells, leukocytes, endothelial cells, and vascular smooth muscle cells.
